AstraZeneca LP has recently introduced two new drug candidates to treat pulmonary hypertension (PH) in South Africa. These drugs have not received approval for the market but are expected to generate substantial revenue as a by-product of the company’s acquisition of a major drug in South Africa. The drugs are:

Indications

Contraindications

Hypersensitivity to tadalafil or similar drugs.

Special Precautions

Patients should avoid using tadalafil; avoid prolonged use; blood pressure monitoring (e.g., pulse, blood pressure, etc.), nitrate drug, or nitroglycerin, or alpha-blocker drugs; high cholesterol or triglycerides; and use alpha-blockers or calcium channel blockers in patients with coronary artery disease. Patients with a history of cardiovascular disease, a recent stroke, sudden cardiac death, or heart attack, taking beta-blockers, including sildenafil, or concomitant use of these drugs should be carefully followed.

Pregnancy and Breastfeeding

Consumption of tadalafil during pregnancy is contraindicated in this population; use in pregnancy is not recommended. It is not known whether the drug is excreted in human breast milk; it is unlikely to harm the developing baby.

Adverse Reactions

• Theophylline, used to treat asthma

• Antibiotics:- Clarithromycin, Erythromycin and Telithromycin

• Anti-fungals such as itraconazole and ketoconazole

• Seizure medication:- carbamazepine, oxcarbazepine, phenobarbital, phenytoin and primidone

• Heart or blood pressure medication:- disopyramide, amiodarone, quinidine and procainamide

• Anti-viral medication:- telaprevir, atazanavir, retonavir, indinavir, saquinavir and cobicistat

• Recreational drugs:- amyl nitrate (poppers)

Do not take Tadalafil if you:

• Tadalafil should never be used with fratterine orIts’ Itchy Nitrates such as nitrate-basedrique or KONV, which can increase the total dosage.

• Men with kidney problems or a history of seizures should not take Tadalafil.
